Possibly yes. It's just hard to say at the moment, what kinds of tests SpaceX will do and in what order. but it's safe to say, they will start with small hops as always. Then go to 5000, 50000 and then above Karman Line. Probably all without Super Heavy and then they will do the same with Super Heavy alone. After that, they will start combining the two.

Is Starship going to do a sub-orbital test flight where it leaves the atmosphere before Super Heavy is ready? Might they do that to test Starship's viability as a point to point transportation vehicle for future passenger flights between cities on Earth?
